Abstract
Objective To study the cooperating effect and the underlying mechanism of endostar(ES) with radiation therapy(RT) in Lewis lung cancer mice.Methods The Lewis lung cancer xenograft model was established by injecting Lewis lung cancer into C57BL/6 mice.The 32 tumor-bearing mice were randomly divided into 4 groups: ES group,RT group,ES combination with RT(ES + RT) group,and control group which was injected with normal sodium chloride solution.The expression of hypoxia inducible factor 1α(HIF-1α),aquaporin-1(AQP1) and vascular endothelial growth factor(VEGF) in Lewis lung cancer mice was tested using immunohistochemistry.The tumor killing effect of RT was also analyzed.Results ①Compared with the control group,the expression of the three proteins were significantly decreased in ES + RT group(P 0.05).As compared with the ES + RT group,the expression of the proteins increased significantly in the RT group,ES group and control group(P 0.05).②HIF-1a showed a positive correlation with VEGF protein(r = 0.504,P 0.05),and the expression of AQP1 decreased along with the descended expression of HIF-1α and VEGF(P 0.01).Conclusion It is demonstrated that treatment with ES + RT results in reduced expression of HIF-1α,AQP1 and VEGF proteins in lung cancer bearing mice.The 3 proteins are positively correlated in the development of lung cancer,so that the treatment in combination with ES could enhance the tumor killing effect of radiation.
