Effect of Cyanin in Protecting Cultured Müller Cells Under High Glucose and High Insulin Condition

Abstract

Objective:To evaluate the activity of cultured retinal Muller cells under high glucose and high insulin condition,and study the effects of cyanin on Muller cells' activity.Methods:Muller cells from rat retina were collected and cultured in vitro,and were divided into three groups as high insulin,low insulin and control groups,according to the treatment of 50 mmol/L glucose plus different concentrations of insulin(3 kU/L,3 U/L,and 0 U/L insulin) respectively for 24 hours.Different concentrations of cyanin(121.1,22.8,and 0 μmol/L) were added to the medium in these groups.After 48 hours incubation,the activity of Muller cells was evaluated by MTT method.Results:After high insulin treatment,the activity of Muller cells decreased dramatically as compared with that of the control group(P0.05).After treated with 121.1 μmol/L cyanin,Muller cells were activated as compared with those without cyanin treatment(P0.05),and showed no significant difference from the control group(P0.05).However,cyanin of low concentration did not have the similar effect(P0.05).Conclusion:High concentration of insulin could decrease the activity of Muller cells in a short time.After treatment of cyanin,the activity of Muller cells in high insulin groups could recover.
