Abstract

Objective To explore the effect of epigallocatechin-3-gallate(EGCG) on the proliferation of osteosarcoma cell and its mechanism.Methods Human osteosarcoma cell line SOSP9607 was cultured with or without EGCG.Then the cell growth curve was drawn.Apoptosis analysis was finished by Hoechst staining.The changes in expressions of survivin mRNA and protein were detected by RT-PCR and Western blot.Results EGCG could markedly inhibit the proliferation of the osteosarcoma cell SOSP9607.The cell apoptosis became obvious 48 hours after the action of EGCG at a dose of 60 μg/mL,with an apoptotic rate of 16.3%(P 0.01).Expressions of survivin mRNA and protein on the osteosarcoma cell were simultaneously reduced.Conclusion EGCG can inhibit the proliferation of the human osteosarcoma cell and induce its apoptosis,which may be associated with suppressing the survivin expression.
