Study on the establishment of siRNA eukaryotic expression vectors of recombinant targeting gene TGF-βIIR and its transfection into human T cells

AIM:To design and establish siRNA eukaryotic expression vectors of recombinant targeting gene TGF-βⅡR,and to transfect them into T cell.METHODS:Recombinant was designed and established by targeting gene TGF-βⅡR based on TGF-βⅡR cDNA sequence. Three pairs of oligonucleotides were synthesized and inserted into plasmid pSilencer 3.1H1 to generate siRNA eukaryotic expression vectors.Then, the plasmids were transfected into human T cells and the results were identified with RT-PCR and Western blot.RESULTS:The recombinant vectors were established successfully and the expression of TGF-βⅡR of T cell which was transfected with the siRNA1 was inhibited obviously.CONCLUSION:siRNA eukaryotic expression vectors of recombinant targeting gene TGF-βⅡR has been established and it has been transfected into T cell successfully.
