Detection of Her Oncogene by Fluorescence In Situ Hybridization in Breast Cancer

Abstract

Objective:To detect the expression of Her2 protein by immunohistochemistry and Her2 gene amplification by fluorescence in situ hybridization(FISH) in breast cancer and evaluate the clinical significance.Methods:The expression of Her2 protein and Her2 gene amplification in 50 cases of breast cancer were detected by immunohistochemical staining and FISH,respectively.Results:Of 50 cases,the expression of Her2 protein were positive in 21 cases and strongly positive in 29 cases,17 Her2 gene amplification existed in 21 cases(++),28 Her2 gene amplification existed in 29 cases(+++),45 Her2 gene amplification in 50 cases.No significant difference was found in the results between these 2 methods.Conclusion:The immunohistochemistry can be used in initial screening of whether select Herceptin in clinical therapy or not.In the cases with positive and strongly positive expression of Her2 protein,Her2 gene amplification should be detected.
