Abstract
Objective: To investigate the role of activated nuclear factor-kappa B(NF-κB) in intestinal mucosal barrier injury of rats with obstructive jaundice.Methods: Thirty male Wistar rats weighing 230-260 g were randomly divided into three groups.They were sham operation group(SH)(n=10),obstructive jaundice group(OJ)(n=10) and the treatment group of OJ treated with pyrrolidine dithiocarbamate(OJ + PDTC)(n=10).The animal model of obstructive jaundice was established. Three weeks after operation,inferior vena cava was put into death in batch and then the tissues of pancreas and the terminal ileum were taken,detecting the changes of plasma endotoxin,D-lactate,TNF-α,IL-6 and bacterial translocation,observing the morphological changes of intestinal lumen mucosa in each one of groups and detecting activated NF-κB in intestinal mucosa with immunohistochemistry S-P method.Results: The bacterial translocation,plasma endotoxin,D-lactate,TNF-α and IL-6 level in OJ group increased significantly compared with SH group and there were significant differences between them(P0.05).The bacterial translocation,plasma endotoxin and D-lactate in OJ + PDTC group decreased significantly compared with OJ group and there were significant differences between them(P 0.05).Few activated NF-κB was expressed in intestinal mucosa of SH group while more activated NF-kB was expressed in OJ group and there were significant differences between them(P0.05).And activatied NF-κB in OJ + PDTC group was significantly fewer than that of OJ group and there were significant differences between them(P0.05).Conclusion: Activation of NF-κB plays an important regulating role in intestinal mucosal barrier injury of obstructive jaundice.
