Abstract
Objective:To investigate the expression and role of TNF-α on rat renal graft administered with ischemic preconditioning.Methods:Thirty-six male SD rats were served as donors and recipients.Animals were randomly divided into the following 3 group:the sham-operated group(group A),common transplantation group(group B)and treatment group(group C).Animals in A group were subjected to exploratory laparotomy.Group B received orthotopic homotransplantations.IPC in C group was administered with a 15 minutes ischemic followed by a 10 minutes reperfusion.The levels of Scr,BUN,the degree of ischemic reperfusion injury of kidney graft were assessed.TNF-α levels in serum and graft tissue were detected by RT-PCR and enzyme-linked immunosorbent assay(ELISA),respectively.Results:The degree of renal graft tubules injury in C group was significantly lower compared with the Group B(P0.01)as well as the transcription and expression of TNF-α(P0.01)after 24 hours of transplant.The levels of Scr,BUN were elevated than the sham-operated group,whereas no significant difference between Group C and Group B(P0.05).Conclusions:One cycle schedule of preconditioning attenuated rats renal graft ischemia reperfusion injury in early phase.The inhibition of expression of TNF-α may play key role for the protection.
