Abstract
Substances such as phenols,pigments in plant tissues affect their RAPD results.In this study,the effect of PVP,Proteinase K,RNase A,the extraction and purification processes on the quality of DNA extracted from sugarcane and their RAPD results were studied.It was found that:(1)the quality of extracted DNA was directly related to the treatments of RNase A and PVP,but not significantly to the extraction processes of CTAB or SDS,or to the treatment of Proteinase K;(2)the RAPD results did not show any difference when there exist little RNA and proteins in the template DNA.The pigments and the template DNA degradation were the main factors that disturb the RAPD results.An effective process for extracting and purifying total DNA of plants was proposed.
