Abstract
Objective: To extract ginkgo polysaccharide from Ginkgo biloba leaves,and to study the protective effect of ginkgo polysaccharides on carbon tetrachloride(CCl 4) induced acute liver injury,and to explore the possible mechanism of them.Methods: 60 Kunming mice were divided into 6 groups randomly(n = 10),respectivley for the control group,model group,bifendate group,ginkgo polysaccharide group with low,medium,high dose.Blank group and model group with saline ig,Ginkgo polysaccharide low,medium and high dose group and bifendate of group were given ginkgo polysaccharide(50,100,200 mg·kg-1) and bifendate(100 mg·kg-1) ig,administered continuously 14 d,1 times / d.Prepare model of acute liver injury in mice with CCl 4.Observe and compare the liver histological changes by the electron microscope.Determine aspartate aminotransferase(AST) and alanine aminotransferase(ALT) in serum superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px),malondialdehyde(MDA) content,NO,TNF-alpha content in liver tissue.Results: Ginkgo polysaccharides can reduce the ALT,AST content in serum of mice siginicantly induced by CCl 4(P 0.01,compared with the model group,the same below),And can increase significantly SOD,GSH-Px content(P 0.01),decrease MDA content(P 0.01),and reduce the NO,TNF-α content(P 0.01).Conclusion: Ginkgo polysaccharides can effectively protect CCl 4-induced liver injury in mice which may be related to free radical scavenging capacity.
