Dynamic observation of the DEXTER culture of cord CD34~+ cells in vitro

Abstract

Objective To observe dynamically the DEXTER culture of cord CD34 + cells in vitro . Methods CD34 + cord cells were separated by MACS system from 9 fresh samples. Stromal cells in umbilical blood were cultured by using DEXTER culture system. After adding the different cytokine(s)individually, the number of cord stromal cell colony was counted and the morphology of the cord stromal cells in culture was observed respectively at different time points. Results SCF+BFGF, SCF could promote the growth of cord stromal cell colony. The biological properties of cord stromal cells were different from those of bone marrow stromal cells at many aspects such as growth condition, conformation, cell morphology, etc . Conclusion Cord CD34 + cells with the aid of some hematopoietic growth factors have promoting effects on the expansion of cord stromal cells and may become a new resource of stromal cells.
