Advances in the therapeutic monoclonal antibodies and the antitumor applications of their immunoconjugates

Abstract

At present,more than a quarter of biotech drugs are monoclonal antibodies.The therapeutic antibodies represent one of the fastest growing areas of the biopharmaceutical industry.They have been used to treat human diseases,such as cancer,transplant rejection,autoimmune disease,cardiovascular disease,and antiviral prophylaxis.Mouse monoclonal antibody has greatly impacted the development of research on human diseases,but which has serious shortage in the treatment of diseases.Now,humannization of monoclonal antibody has attracted significant research.Immunoconjugate agents can be obtained by coupling the radioactive materials,chemotherapy agents and biotoxin with monoclonal antibody.The coupled agents produce antitumor effect by directing monoclonal antibody to the cell surface that expresses corresponding antigen.The coupled agents also decrease the damage to normal tissues.This article reviewed the recent progresses in the studies of monoclonal antibody humanization and its antitumor applications.
