Preparation and Evaluation of Specific Yolk Antibodies Against H.Pylori

Objective To find an effective way to prepare the specific Yolk antibodies against Helicobacter pylori(HP) and to evaluate the antibody titer.Methods Six 31-week-old Hy-Line laying hens were immunized with inactivated HP antigen and its ultrasonic HP fragment antigen combined with either Freund's complete adjuvant or Freund's incomplete adjuvant.The second immunization was performed after 2 weeks,and the third immunization was taken after 4 weeks.The eggs were collected after the first immunization and the specific Yolk antibodies anti-HP were prepared.The immune response was detected by IHA and the bioactivity and titer of anti-HP IgY were evaluated.Results Two weeks after the first immunization,the titer of the antibodies reached 1∶256.Three weeks after the first immunization,the titer reached 1∶1 024.Four weeks after the first immunization,the titer reached 1∶8 192 and was maintained at this level for more than 11 weeks,then the titer dropped slightly.After 26 weeks,the titer still was maintained at a high level.In addition,IgY-HP restrained the growth of HP.Conclusion Booster immunization is a good way to acquire the effective antibodies.
