Content determination of quercetin and kaempferol in Hedotis diffusa Willd by HPLC

Abstract

Objective:To determine the content of quercetin and kaempferol in Hedyotis difusa Willd.Methods:The analysis was performed on an HPLC C18 column(4.6 mm×250 mm,5um)at the detection wavelength of 370 nm.The mobile phase consisted of second nitriles-methanol-0.2% phosphoric acid(10:45:60) with a flow rate of 1 mL/min.Results: The calibration curve was linear in the range of 4.20~25.20ug(r=0.9999) of quercetin.And in the range of 0.97~11.64ug(r=0.9997) of kaempferol.The average recovery was 100.48% with RSD 1.01%(n=3) of quercetin and 97.85% with RSD 1.36%(n=3) of kaempferol.Conclusion: The method is simple,rapid and accurate for the determination of quercetin and kaempferol in Hedyotis diffusa Willd,which could be used for the quality control of Hedyotis difusa Willd.
