Optimization on fermentation medium for ε-polylysine by response surface methodology

Abstract

Plackett-Burman design and response surface method were applied to obtain optimum media fore-Polylysine production by streptomyces albus KD-11.Firstly,Plackett-Burman design was used to evaluate the remarkable variables which were relevant to the production of e-Polylysine.Then the greatest response area was studied by steepest ascent approach design.Finally,the optimum medium was selected using the central composite design and Response Surface Methodology.The results showed that both ammonium sulfate and glucose have significant influence one-Polylysine yield which optimum concentrations were 33.196 g/L,8.572 g/L respectively and the experimental e-Polylysine yield could reach 2.491±0.124 g/L on optimized medium,which agreed with the predicted value of 2.543 139 g/L and was increased by 63.6%.
