Effects of diethylstilbestrol on testicle development of fetal mice

Abstract

The effects of diethylstilbestrol(DES) on testicle development of fetal mouse were studied in the experiment.Forty female mice were divided into 4 groups randomly,control group and 3 experimental groups.DES was injected hypodermically from 10 to 18 days at dosage of 2,4 and 8 μg for each mouse per day after pregnancy while the control group was administrated with saline hypodermically.After 2 hours of the last drug administration,the testicle of embryo was collected and observed in the HE stain.Expression of proliferating cell nuclear antigen(PCNA) in testicle tissue was detected with immunohistochemistry.The results were showed as follows: 1) Development of testicle was close to the control group in 2 g group.2) There was decrease in amount of gonocyte and interstitial cell in 4 g group.3) Nucleolus of gonocyte was not clear,amount of interstitial cell was reduced,sertoli cell was dissolved in 8 g group.4) Content of PCNA was decreased as dosage of DES increased(P0.05).All above indicated that the DES could inhibit the proliferation of orchic cells in embryo,then influenced the testicle development,damaged the testicle structure.
