Abstract

Objective To explore the influence of different blood serum concentration and different in vitro culture time upon the levels of type Ⅰ and Ⅲ collagen and their ratio of L929 cell line of the mouse fibroblast. Methods L929 cell line of the mouse fibroblast was cultured in vitro, and ABC-ELISA was used to measure the concentrations of type Ⅰ and Ⅲ collagen and their ratio. Results Except 0% serum concentration group which type Ⅰ and Ⅲ collagen displayed no significant changes but only a slightly increasing tendency, type Ⅰ and Ⅲ collagen in other serum concentration groups displayed an increasing state and increased most obviously from the 6th to 8th day.The ratio of type Ⅰ to Ⅲ changed differently in different concentrations of blood serum for different growth time.In 20% and 40% serum concentration groups, the ratio of type Ⅰ to Ⅲ increased gradually with the cell growth time.In other serum concentration groups, the ratio of type Ⅰ to Ⅲ did not increase obviously with the cell growth time. Conclusions Different blood serum concentration and different in vitro culture time of L929 cell have a certain influence upon the type Ⅰ and Ⅲcollagen and their ratio. This provides experimental data for future improvement of in vitro cell culture environment.
