The frequency of the ABCA1 R219K polymorphism in Patients with Coronary Heart Disease and Type 2 Diabetes Mellitus

Abstract

Objective To evaluate the frequency of R219K polymorphism of ABCA1 gene on blood lipids and its relationship with coronary disease(CHD) and melitus diabetes(DM) in Northern Han ethnic group.Methods DNA was extracted with meliorated iodonationnatrium(NaI) method,the genotypes of R219K of ABCA1 gene were detected with polymerase chain reaction-restrictive fragment length polymorphism(PCR-RFLP).Results The frequency of the ABCA1 R219K genotypes(RR,RK,KK),R and K allele is 28.5%,48.6%,22.9%,52.8%,47.2% in controls.29.6%,63.9%,6.5%.61.5%,38.5% in DM group,44.7%,44.7%,10.6%,67.0%,33.0% in CHD group,30.5%,61.9%,7.6%,61.4%,38.6% in DM+CHD group.Conclusion R219K polymorphism of ABCA1 gene has a great effect on serum HDL-C level,which is higher in subjects carrying the K allele than R gene carrier population.The contribution of R219K polymorphism of ABCA1 gene is significant difference among the DM,CHD,and DM+CHD group.K allele is associated with CHD accidents.
