Abstract

Objective:To investigate the effect of Hugu Capsule on proliferation and differentiation in primary osteoblasts.Methods:Bone cells were obtained by enzyme digestion from the segregated neonatal SD rat skull.The effects of Hugu Capsule on the proliferation and differentiation of rat osteoblasts were evaluated by the MTT method,measuring the activity of alkaline phosphatase,respectively.Runx2 expression was analyzed by real—time fluorescent quantitative reverse transcriptase polymerase chain reaction(qRT—PCR) using SYBA Green I.Results:With the concentration of 0.025-2.5 μ g/mL,Hugu Capsule had significant effect on the ROB's proliferation.The ALP activity was significantly increased at the concentration from 0.025 μg/mL to 25 μ g/mL.With the concentration of 25 μ g/mL,Hugu Capsule can increase the Runx2mRNA level.Conclusions:Hugu Capsule can stimulate the proliferation and differentiation of osteoblasts in vitro,and the reason may be connected with the up regulation of Runx2 mRNA level.
