Abstract

Objective To discuss the inhibitory effects of suramin on the proliferation of cultured Tenon's capsule fibroblasts.Methods The fibroblasts were isolated from glaucoma patients Tenon's capsule,cultured and subcultrued in vitro.The proliferation of fibroblasts was detected by MTT method after incubation with different concentration of suramin with different time,while the cell cycle was analyzed by FCM.Results MTT assay showed that suramin started to reduce fibroblasts proliferation at the concentration higher or equal to 500 mg·L~-1?400 mg·L~-1?300 mg·L~-1 after 24 hours?48 hours?72 hours of incubation with suramin respectively. FCM revealed that fibroblasts in G_2/M phase increased 15.8% compared to control group.Conclusion It was demonstrated that suramin could inhibit the proliferation of Tenon's capsule fibroblasts in vitro,as dose- and time-dependent manner.These findings suggested that suramin may be beneficial to patients with ocular fibroliferative disorders.
