Inhibitory effect of quercetin on human Tenon’s capsule fibroblasts

Abstract

Objective To investigate the inhibitory effect of quercetin(QU) on proliferation of human Tenon’s capsule fibroblasts.Methods Human Tenon’s capsule fibroblasts were cultured in vitro and exposed to different concentrations of QU solution,which were 20 μmol·L-1,40 μmol·L-1,60 μmol·L-1,80 μmol·L-1 and 100 μmol·L-1.The proliferation of human Tenon’s capsule fibroblasts was detected by CCK-8 agent,and the shape changes of cell were observed with light microscope.Results After Cells were treated by 50 μmol·L-1 QU for 48 hours,we observed that cells were shrunk and aggreagted to nuclear membrane edge.After cells were treated with 100 μmol/L QU for 48 hours,the proliferation of human Tenon’s capsule fibroblasts was completely inhibited,and apoptotic bodies were seen.We could observe apoptotic body increased and the proliferation of human Tenon’s capsule fibroblasts was completely inhibited.Conclusion QU can inhibit the proliferation of human Tenon’s capsule fibroblasts in vitro and show a dose-time dependent tendency.It is potentially useful for preventing scar formation in the glaucoma filtration surgery.
