Optimization of extraction conditions for antioxidant and free radical scavenging substance production from pogadatree flower

Abstract

The present study was to optimize the extraction conditions that give maximum free radical scavenging substances from pogadatree flower(Sophora japonica L.). The results showed that the pogadatree flower extraction with 70% ethanol provided the highest free radical scavenging substance,total phenol and total flavonoids content. The optimal extraction temperature for superoxide anion scavenging substance was identified as 45℃,and other free radical scavenging substances were determined as 50℃. The rate of material and solvent for most efficient extraction of total antioxidant activity,DPPH,hydroxyl radical-scavenging substance was 1∶20,and the remainders was 1∶15. Twice extraction of pogadatree flower showed about 83%~95% of the total free radical scavenging substance,phenol and flavonoids content. The results of correlation analysis showed that total antioxidant activity and DPPH free radical mainly came from phenol and flavonoids.
