Abstract
Objective To discuss the mechanism of the protective effect of FMCL on apoptosis of PC12 cells induced by H_2O_2.Methods Based on the model of apoptosis of PC12 cells induced by H_2O_2,apoptosis rate was analyzed by flow cytometer.The expression of mRNA and protein were tested by RT-PCR and Western blot.Results The apoptosis rate was decreased in the FMCL treatment group.The mRNA and protein expression of Bcl-2 were increased and that of Bax was decreased(P0.05 or P0.01).Thus the ratio of Bcl-2/ Bax increased.The activity fragment of caspase-3P20 was decreased(P0.05).Conclusion FMCL can prevent PC12 cells against H_2O_2 oxidation damage.The mechanism of protection is likely related to increasing the ratio of Bcl-2/ Bax so that can decrease the activation of caspase-3.
