Abstract
Objective To study the effect of Schisandrin(Sch B) on apoptosis of PC12 cells induced by H2O2 and its mechanism.Methods Based on the model of apoptosis of PC12 cells induced by H2O2,cell survival rate was evaluated by MTT assay,and apoptosis was analyzed by Hoechst33258 fluorescence staining and flow cytometer.The DCFH-DA fluorescence staining was used to determine the level of reactive oxygen species(ROS) in PC12 cells.The expression of mRNA and protein for p53 gene were tested by real time PCR and Western blot.Results Treatment with different concentration of Sch B(5~40 μmol/L) for 48 h increased the survival rate of PC12 cells in a dose-dependent fashion,Sch B prevented the PC12 cells nuclei from shrinkage,condensation and cleavage induced by H2O2.Sch B decreased the level of ROS and the expression of p53 mRNA and p53 protein in the PC12 cells.Conclusions Sch B can prevent apoptosis of PC12 cells by H2O2 in a dosedependent fashion,which is likely related to antioxidation and decreasing p53 gene expression.
