In Vitro Study of the Reversal Effect of Tetrandrine Combining with Celecoxibon Multidrug Resistance in Leukemia Cell Line

Abstract

Objective: To investgate whether tetrandrine combining with celecoxib can reverse multidrug resistance in leukemia cell line K562 and explore its possible mechanism.Methods: Cytotoxic effect of tetrandrine alone and tetrandrine combining with celecoxib on K562 cell line was detected using MTT colorimetry;apoptosis and intracelular drug concentration were determined by flow cytometry(FCM);the expression of COX-2/COX-1 was detected using RT-PCR.Result: Compared with other groups,combination of tetrandrine and celecoxib had stronger inhibition effect on K562 cell line and higher intracelular drug concentration and apoptisis percentage(17.67%),however,the expression of COX-2/COX-1 was lower than other groups.Conclusin: Tetrandrine combining with celecoxib can reverse the multidrug resistance,the underlying mechanism may attribute to increaseing intracelular drug concentration、induction of apoptosis and down-regulation of mRNA expression of COX-2/COX-1.
