Extraction and Determination of Total Flavonoids from Puerarin

Abstract

Using ethanol to extract the total flavonoids from the pueraria,the effects of extraction temperature,extraction time,ethanol concentration and solid-liquid ratio on total flavonoid yield were investigated,the optimum conditions of the extraction process were determined,and the content of the flavonoids was determined by UV light spectrophotometry method.The experimental results showed that the yield of the total flavonoids was 3.3% when extraction temperature was 70℃,solid-liquid ratio was 1∶20,ethanol volume fraction was 80%,and the extraction time was 120 min.The average recovery rats of 97.5% and small standard deviation and variation coefficient showed the precision of the detection method and good reproducibility.It is an effective method to detecte the content of the total flavonoids in pueraria.
