Abstract
【Objective】 The paper mainly uses dual-wave length spectrophotometry method to study the determinnation of amylose and amylopectin in grain and bean.【Method】Based on the absorption spectra of the two complexes,the amylose-wave length of 495/565 nm、and amylopectin wave length of 530/654 nm were obtained as the measuring wave length.According to the linear regression equationes,the paper can determine the content of amylose and amylopectin in sample solution.【Result】 The result shows that amylose concentration is between 0~80 mg/L and amylopectin concentration is between 0~400 mg/L.【Conclusion】The content of amylose and amylopectin determined by the method is accurate,because it is not influenced by the reagent and the error which is caused by the absorption cell.So that it is most suitable to determine amyloy and amylopectin and the results of the experiments is most satisfactory.
