Reversal Effect of Sophoridine on B7-H1-mediated Multidrug Resistance in Human Gastric Cancer Cells

Abstract

Objective:To investigate the reversal effect and mechanism of sophoridine(SRI) on human gastric cancer multidrug resistance(MDR) in SGC7901/DDP cells.Methods: Human gastric cancer cells SGC7901 and MDR SGC7901/DDP w ere cultured conventionally.After treatment w ith SRI for 48h,the sensitivity of SGC7901/DDP cells to chemotherapy drugs w as evaluated by the MTT assay,the expression of B7-H1 w as evaluated by Western blot,and the effects of SRI on apoptosis and cell cycle w ere evaluated by flow cytometry(FCM).Results:The sensitivity of DDP in SGC7901/DDP cells w as much low er than that in SGC7901 cells(P 0.01).After treatment w ith SRI for 48h,cell proliferation w as inhibited,apoptosis rate w as increased,and the expression of B7-H1 w as dow n-regulated in human gastric cancer multidrug resistance SGC7901/DDP cells(P 0.05).Conclusion:SRI can reverse MDR in human gastric cancer cells SGC7901/DDP.The possible mechanism is related w ith dow n-regulating B7-H1 expression,inhibiting cell proliferation and inducing cell-apoptosis.
