Abstract
Objective To investigate the effect of rosuvastatin on angiogenesis of myocardium in diabetic rats model with myocardial infarction(MI).Methods Rats with diabetes were randomly assigned to Sham operation group(group A),MI group(group B),rosuvastatin group(group C).Echocardiographic examination was conducted.Masson's staining was used to detect the level of fibrosis of myocardium.Immunohistochemistry was used to determine the expressions of VEGF and CD31.The expression of VEGF mRNA was detected by RT-PCR.Results Compared with group A,the left ventricular ejection fraction was significantly lower, the left ventricular internal diameter at end-systole(or end-diastole) was much longer, and the level of fibrosis of myocardium was significantly increased in group B(P 0.05 for all).Compared with group B,the left ventricular ejection fraction was significantly higher, the left ventricular internal diameter at end-systole(or end-diastole) was much shorter, the level of fibrosis of myocardium was significantly decreased, the expressions of VEGF and CD31 of myocardium were significantly increased, and the expression of VEGF mRNA was significantly elevated in group C(P 0.05 for all). Conclusion Rosuvastatin effectively increased angiogenesis of myocardium in diabetic rats with myocardial infarction, and thus may play a crucial role in the prevention of cardiac dysfunction after myocardial infarction, possiblely resulting from the up-regulation of VEGF expression.
