Abstract

[Objective]The study aimed to screen the best medium for bud induction,proliferation,rooting and the matrix for training plantlets of Bretschneidera sinensis.[Method]With the embryo bud of aseptic budding of B.sinensis seeds as explant,and MS as basic medium, cytokinins BA,auxin IBA and NAA with different concn.were added to conduct bud induction culture;with agar,sucrose,NAA,BA with different concn.as factors to do the orthogonal experimental design for multiplication cultivation of subculture,and the IBA and NAA with different concn.were added for rooting culture.[Result]The optimal medium for B.sinensis bud induction was MS + BA 1.0 mg/L + NAA 0.1 mg/L,with the budding percentage as highest as 71.34%and the bud height of 5.30 cm;the best enrichment medium was MS + BA 2.0 mg/L + NAA 0.1 mg/L + agar 1.0%+ sucrose 2.5%,with embryo multiplication coefficient of 3.6 and the bud height of 3.1 cm. The optimum rooting medium was MS + IBA 3.0 mg/L + agar 0.8%+ sucrose 3.0%,with rooting rate of 89%;the optimum matrix for training plantlet of B.sinensis tube seedling was vermiculite 25%+ pearl 25%+ riversand 50%,with transplanting survival rate up to 65%.[Conclusion]The technical system of rapid propagation with tissue culture of B.sinensis was established.
