【Objective】To investigate the relationship between the expression of matrix metalloproteinases(MMP1 and MMP-2) with local invasion and lymphoid metastasis of laryngeal carcinoma.【Methods】Expression of MMP-1 and MMP-2 in 40 cases of laryngeal carcinoma and 10 cases of vocal cord polyp were detected by immunohistochemistry,and their relationship with stage,lymphoid metastasis and clinical data were analyzed.Expression of MMP-1 and MMP-2 both in Hep-2 cells and in xenografted nude mice were also detected by immunohistochemistry.【Results】The positive rate of MMP-1 and MMP-2 in laryngeal carcinoma was 76.7%(23/30) and 80%(24/30) respectively,which were significantly higher than those in vocal cord polyp(P 0.05).The positive rate of MMP1 and MMP-2 in laryngeal carcinoma with T3~4 were both significantly higher than those with T1~2(P 0.05);and there was a statistical significant difference between the positive rate of MMP-1 with and without lymphoid metastasis(P 0.05),the same as MMP-2.There is a positive correlation between MMP-1 and MMP-2,the correlation coefficient is 0.694(P 0.05).Aging and sender had no affect on expression of MMP-1 or MMP-2(P 0.05).The MMP-1 positive cells and MMP-2 positive cells were more than 75% in Hep-2 cells smear.A high expression of MMP-1 and MMP-2 were seen in cancer nest of xenografted nude mice.【Conclusions】There is a high expression of MMP-1 and MMP-2 in laryngeal carcinoma cells,and the expression has a close correlation with local invasion and lymphoid metastasis of laryngeal carcinoma.Detection of MMP-1 and MMP-2 can be used as a index to predict malignant degree,invasive ability and lymphoid metastasis,which has important clinical significance.
