Abstract
To compare the transfection efficiency and proliferation abilities of canine bone marrow mesenchymal stem cells(BMSCs)after transfection of enhanced green fluorescent protein(EGFP)gene by adenoviral vector and lentiviral vector which mediated,the method of whole bone marrow culture was used to isolate canine BMSCs.Canine BMSCs were exposed to different multiplicity of infection(MOI)of adenoviral vector and lentiviral vector.Transfection efficiency was calculated after 72h,and proliferation ability of BMSCs after transfection was detected by MTT reduction assay.The results showed that with the increase of MOI,the EGFP positive rate of BMSCs transfected by each kind of vectors gradually increased,while the proliferation ability of BMSCs after transfection was decreased.When MOI was 25,50,100,200and 400,the transfection efficiency of adenoviral vector was 11%,17%,32%,51% and 65%,respectively,and the transfection efficiency of lentiviral vector was 48%,70%,91%,98% and 99%,respectively.MTT assay showed that proliferation ability of BMSCs was significantly affected by adenoviral vector(MOI=200)and lentiviral vector(MOI=400)in contrast with control group(P0.05).The results indicated that the efficiency of lentivirus vectors was significantly higher than that of adenoviral vectors at the safety MOI.
