Association of CD19 gene polymorphism with systemic lupus erythematosus in Chinese

Abstract

Objective To investigate the possible association of a single nucleotide polymorphism(SNP)at position 705 in exon 4 of CD19 gene(CD19 gene 705) in Southern Chinese patients with systemic lupus erythematosus(SLE) and lupus nephritis(LN).Methods One hundred and three patients(13 males and 90 females,62 cases with LN) fulfilled the American College of Rheumatology 1982 revised criteria for SLE and 110 healthy ethnically matched controls(21 males and 89 females),all from the south of China,consented to participate in the study.Blood samples for DNA extraction from all subjects were collected in EDTA tube.Genomic DNA was extracted according to the standard isolation procedures.CD19 gene 705 polymorphism was typed by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).Results CD19 gene 705 polymorphism was found widely in Southern Chinese.All genotype frequencies of CD19 gene 705 were in Hardy-Weinberg equilibrium.There were not significant differences of CD19 gene 705 genotype,allele and phenotype frequencies between the SLE patients and the controls(P0.05).Similarly,no significant differences of CD19 gene 705 genotype,allele and phenotype frequencies were seen between the male,female SLE and the controls,the SLE with lupus nephritis(LN) and the controls,the SLE with LN and the the SLE without LN. Conclusion CD19 gene 705 polymorphism does not appear to be associated with susceptibility to SLE and LN in Southern Chinese.
