Establishment and judgement of the focal ischemia reperfusion model in rats

Abstract

Objective:To study the influence of reperfusion after ischemia for different time on cerebral infarction volume and neurological function of the focal ischemia reperfusion model in rats. Methods: The 24 rats were randomly divided into three groups: sham operation group; ischemia for 1 hour reperfusion group; ischemia for 2 hours reperfusion group;8 rats in each group. The string-fastening method was used to establish focal ischemia reperfusion models in rats, sham operation rats, the string was only inserted in CCA. The animals were reperfused on 1 h or 2 h after ischeme respectively on the basis of the different group. Their neurological deficits were evaluated and infarct size was analyzed with 2, 3, 5-triphenyltetrazolium chloride (TTC) staining. Results: As compared to ischemia for 1 hour reperfusion group, 2 h group increased significantly neurological deficits scoring of 2h and 6h point after reperfusion (P0.05). There were no obvious differences between 1 h and 2 h group in both scoring and the volumes of cerebral infarction after 24 h reperfusion (P0.05). Conclusion: It’s reasonable for the model to choice ischemia reperfusion for 1 hour.
