Cloning of a Human Arsenic Resistance Related Gene (hARRG) cDNA and Its Expression in E.coli

Abstract

A novel human full length cDNA was cloned from a fetal brain cDNA library through a high throughput cDNA sequencing project. It showed 88 % identity in nucleic acid sequence to asr 2 of hamster. It was named human arsenite resistance related gene ( hARRG ). Moverover hARRG was almost completely identity to ARS 2 (GenBank No. AF082871) except that hARRG was 253 bp shorter than ARS 2. It seems not a full length gene. A full length hARRG cDNA of 2 999 bp was cloned with Northern blot, electron RACE and 5′ SMART RACE. This cDNA is located in chromosome 7q21.3 by Unigene. It has a integrated ORF, encoding a protein of 788 amino acids, with putative molecular weight of 89.2 ku. The ORF of hARRG was subcloned into reconstructed PET11c plasmid and found expressed in E. coli . with IPTG inducing.
