Abstract
AIM:To obtain specific polyclonal antibodies of ruscogenin by preparing artificial immunogen.METHODS:Immunogen RUS-2HS-BSA and coating antigen RUS-2HS-OVA were prepared by active ester method.The ruscogenin antiserum was produced from New Zealand's rabbits immunized with conjugates RUS-2HS-BSA.Moreover,the ruscogenin antiserum were determined with indirect enzyme-linked immunosorbent assay(ELISA)method.RESULTS:It was found that the titer of antiserum was 1:25 600.Additionally,it was proved that the ideal coating antigen concentration was 1.563 μg·mL-1,the available antiserum dilution was 1∶8 000.CONCLUSION:The artificial immunogen could be synthesized by active ester method to get ruscogenin antiserum.
