Effect of Dexmedetomidine on pulmonary injury induced by early-stage severe acute pancreatitis in rats

Abstract
Objective To investigate whether dexmedetomidine can alter the pulmonary induced by severe acute pancreatitis( SAP) in rats during its early period. Methods Seventy-two adult Sprague-Dawley( SD) rats were randomly divided into nine groups with eight animals in each: sham group,SAP 30min,60min,120min,360min groups and dexmedetomidine treatment 30min,60min,120min,360min groups. The preparation of SAP groups were induced by retrograde injection of 5% sodiun taurocholate into the pancreatic duct of rats. The treatment groups received dexmedetomidine with doses of 1μg/kg injected continually immediately in vena caudalis after the Dex were prepared. Rats were sacrificed at different time point,and lung tissues were obtained for pathological study,dry / wet ratio and MPO examination. The levels of SP-A in serum were measured by ELISA,and the expression of NF-κB of lung tissues by immunohistochemical methods. Results After inducing ASP model,NF-κB activity was strengthened and SP-A in serum were significantly higher,accompanied with gradual increase of dry / wet ratio and MPO activity. In the Dex group,the concentrations of SP-A in serum were significantly lower than SAP group at 60min,120min and 360min( P 0. 05),while the expression of NF-κB in lung tissue is also lower than the SAP group at 120min and 360min( P 0. 05),which means the differences were significant. Conclusion Dexmedetomidine has lung protection effect on acute necrotizing pancreatitis-associated acute lung injury.
