Significance of Fas/FasL expression and its apoptosis mechanism in the choriocarcinoma cell lines

Abstract
Objective To observe the expression of Fas/FasL in choriocarcinoma cell lines(JEG-3 and BeWo) and mechanism of T cell apoptosis induced by co-cultured choriocarcinoma and Jurkat cells.Methods Positive expression of Fas/FasL was detected by immunofluorescence microscopy.The situation of Jurkat cells that co-cultured with choriocarcinoma cells at different time was detected by MTT.The apoptosis of Jurkat cells was detected by flow cytometry when FasL of the choriocarcinoma cell was closed by neutralizing antibody NOK-2 for 24 h.The apoptosis cellular morphology was observed by transmission electronic microscope(TEM).Results Fas and FasL were expressed in both human choriocarcinoma cell lines.Compared with Jurkat cells,the expression of Fas mRNA was lower,while FasL mRNA was higher in choriocarcinoma cell lines with a significant difference(P0.05).Co-cultured with Jurkat cells,choriocarcinoma cells induced Jurkat cell apoptosis(P0.05),with extended culture time,the apoptosis rate of Jurkat cells was notably increased(P0.05).The apoptosis rate of Jurkat cell was decreased,after FasL of the choriocarcinoma cell was closed by neutralizing antibody NOK-2.Conclu-sion Fas and FasL are both expressed in human choriocarcinoma cell lines.The choriocarcinoma cell can resist the apoptosis induced by Fas/FasL system,the higher expression of FasL can inhibit immunologic function,and its lower Fas expression in T cells can evade immunologic destruction.
