Study on Outdoor Rooting in Bag of Virus-free Tissue Cultured Plantlets of Sugarcane

Abstract

In order to improve the outdoor transplanting survival rate of virus-free tissue cultured plantlets of sugarcane,the tissue cultured plantlets of ROC22 were taken to conduct outdoor rooting culture experiment in polypropylene bag,and the contamination rate,the growth status of root and leaf and the transplant survival rate of tissue cultured plantlets in different seasons and under different outdoor light conditions were compared.The results showed that the contamination rate of tissue cultured plantlets rooting in bag outdoor was 3.5% to 4.7% in different culture time,which was lower than that of tissue cultured plantlets rooting in bottle outdoor(8% to 14.5%),and the differences reached extremely significant;the effects of different light conditions and environmental temperatures on rooting rate and root number per plantlet were not remarkable;the high temperature season had a certain affection on leaf growth;light condition affected transplanting survival rate of plantlets,when light condition was 70%,the transplanting survival rate of plantlets were the highest,and the other light conditions all reached significant level.After transplanting,the plantlets that rooting in bag outdoor will grow quickly and strong.So,we can conclude that using polypropylene bag as container to rooting tissue cultured plantlets of sugarcane can improve the quality and the transplanting survival rate of rooted-plantlets.
