Effects of Edaravone on content of cerebra and serum SOD,NO,MDA in rats with focal cerebral ischemia and reperfusion

Abstract
Objective To investigate the effect of Edaravone on focal cerebral ischemia/reperfusion injury. Methods The model of focal cerebral ischemia in rats were established by Suture-occluded method. Rats were randomly divided into sham-operated group, focal cerebral ischemia group and Edaravone-treating group, whose arteries were occlusive for 1 hour. All groups were further divided into 4 sectors according to the reperfusion time (2 h, 6 h, 12 h and 24 h) to test the concentration of brain tissue, serum super oxide dismutase (SOD), nitric oxide (NO) and malondialdehyde (MDA). Results For focal cerebral ischemia group, concentration of brain tissue SOD decreased and serum SOD increased and then decreased. The concentration of brain tissue NO decreased and then increased while serum NO concentration kept increasing. And MDA concentration of brain tissue and serum all increased firstly and then decreased. For Edaravone-treating group, the decrease of SOD was comparatively lower than focal cerebral ischemia group. NO and MDA concentration were all significantly lower than those of in control group. In the 6 h-reperfusion sector, moisture content of brain tissue was also significantly lower than that of in control group (81.91±1.66:76.51±1.98). Conclusion Edaravone may reduce free radical to protect against cerebral ischemia/reperfusion injury.
