Effect of Cigarette Smoke on Expression of IL-1,IL-6 and IL-8 in Lungs

Abstract

Objective To investigate the effect of cigarette smoke on the expression of inflammatory mediators in lungs.Methods A549 cells were exposed to different concentrations of cigarette smoke extract(CSE),which was collected by using dimerhyl sulfoxide(DMSO) as absorbent.The level of IL-1,IL-6 and IL-8 was measured with radioimmunoassay.The animal models of smoking were established though different doses smoking exposure for 12 weeks.Inflammatory mediators in bronchoalveolar lavage fluid(BALF) were detected by radiation immune method.Results In cell experiment,no significant difference was seen in the expression of IL-1 among groups(P0.05).Compared with the control group,the IL-6 expressions in 20%,50%,100% concentrations groups significantly increased(P0.05),the expression of IL-8 decreased(P0.05).In animal experiment,also,no significant difference was seen in the expression of IL-1 among groups(P0.05).The levels of IL-6 and IL-8 in BALF increased significantly in high-dose group,moderate-dose group and low-dose group respectively compared with the control group(P0.05).Conclusion Cigarette smoke exposure can influence the expression of IL-6 and IL-8 in lungs.
