Influence of cigarette smoke on the expression of IL-1,6,8 in rats' lungs and A549 cell

Abstract

OBJECTIVE To investigate the influence of cigarette smoke on the expression of inflammatory mediators in lungs.METHODS The animal model of smoking was established.Each group of rats was given smoking by the respective doses for 12 weeks.IL-1,IL-6 and IL-8 in bronchoalveolar lavage fluid(BALF)were detected by radiation immune method.Human lung adenocarcinoma A549 cells were cultured.Mainstream smoke was collected by using dimethyl sulfoxide(DMSO)as absorbent.A549 cells were incubated with different concentrations of CSE(cigarette smoke extract).After 4 hours the level of IL-1,IL-6 and IL-8 was measured with radioimmunoassay.RESULTS The expression of IL-1 in A549 cells with different concentrations of CSE had no difference with the control cells(P﹥0.05).Compared with the control group,the IL-6 expressions of 20%,50%,100% concentrations increased(P﹤0.05),while the expression of IL-8 was decreased by the CSE(P﹤0.05).The expression of inflammatory mediators in lung of the smoking rat was influenced:For IL-1 in the smoking rat,different dose of smoking had no difference with the control cells(P﹥0.05).The levels of IL-6 and IL-8 in BALF increased significantly in high-dose group,middle-dose group and low-dose group respectively compared to that of Control group(P﹤0.05).CONCLUSION Cigarette smoke could influence the expression of IL-6 and IL-8 in lungs.
