Abstract

[Objective] The research aimed to establish the tissue culture and rapid propagation technology system of big sweet cherry rootstock Gisela.[Method]With the shoot apical meristem of Gisela No.5 as material, the paper studied the tissue culture and rapid propagation technology from axillary buds induction, subculture, rooting and plantlet hardening and transplantation.[Result]The results showed that the optimum medium for axillary buds induction was MS+6-BA 1.00 mg/L +NAA 0.03 mg/L +KT 1.50 mg/L +GA3 0.20 mg/L;1/2 MS+IBA 1.50 mg/L was suitable for the test tube plantlet rooting. The young plantlet had high survival rate when took the 1/2 straw dung and 1/2 garden soil as transplantation substrate.[Conclusion]This study will provide the theoretical and practical basis for rapid production of tissue culture of Gisela No.5.
