Abstract
COX-2 plays an important role in the progress of malignent tumor. The aim of this study is to observe the ability of celecoxib to sensitize five different strains of human malignent cells to radiation in vitro and to prove the possible radiosensitive mechanism. The subtoxic dose of celecoxib was chosen to make the radiosensitive experiments on the human glioma cell lines SHG-44 and 767,the human lung adenocarcinoma cell lines A549,the human cervical carcinoma cell lines HeLa and the human nasopharyngeal carcinoma CNE.The cells were planted into two groups respectively, one was radiation only(R),and the other was radiation combined with 30μmol/L celecoxib(R+ D).Celecoxib's radiosensitization was measured by clongenic assay.Celecoxib displayed radiosensitizing effect in the five strains of malignent cells. The survival curve and the related parameters such as: D0,Dq,SF2 and D0.01in group D+ R decreased significantly from Group R.It is indicated that the subtoxic dose of Celecoxib could enhance radiosensitivity of SHG-44 and 767 cells,A549 cells,HeLa cells and CNE cells in vitro. The radiosensitivity enhencement by the affection of Celecoxib is universal in different malignant tumor cells.
