Effects of propofol on total hepatic ischemia-reperfusion induced lung injury in rats

Abstract

Objective To explore the effects and the possible mechanism of propofol on total hepatic ischemia-reperfusion (I/R)-induced lung injury in rats.Methods Thirty-two rats were equally randomized into 4 groups. The rats in group Ⅰ were given intraperitonelly propofol 50 mg/kg at 10 min before ischemia and normal saline 50 ml/kg at 10 min before reperfusion,those in group Ⅱ normal saline 50 ml/kg at 10 min before ischemia and propofol 50 mg/kg at 10 min before reperfusion,those in group Ⅲ normal saline 50 ml/kg at 10 min before ischemia and at 10 min before reperfusion,and those in group Ⅳ normal saline 50 ml/kg at corresponding time points after sham-operation. One hour after reperfusion,the lung tissues were taken for determination of myeloperoxidase (MPO) activity,malondialdehyde (MDA) content,malondialdehyde (SOD) activity,and weight/dry (W/D) ratio. Pulmonary histomorphology and apoptosis were observed via optical microscope. Results Compared with groups of Ⅰ,Ⅱ,and Ⅳ,W/D ratio,MDA content,MPO activity,apoptotic index were increased,and SOD activity decreased in group Ⅲ(P0.01). Severe lung injury and many inflammatory cells infiltration were observed in group Ⅲ via optical microscope.Conclusion Propofol has protective effect on the lung injury induced by total hepatic I-R,which possibly is mediated by anti-oxidation,anti-inflammation,and anti-apoptosis.
