Abstract
Objective:To Study the effect of brain hippocampus reactive gliosis in the process of occurrence and development of epilepsy and the effect of Topiramate(TPM)on it.Methods: A model of chronic epilepsy was established with pentylenetetrazol.Rats were randomly divided into 3 groups:control group(C group)、PTZ group(P group,P1~10 d、P2~20 d and P3~27 d)and TPM+PTZ group(TP group,TP1~10 d、TP2~20 d and TP3~27 d).The behavior and expression of Glial fibrillary acidic protein(GFAP)by immunohistochemical staining were observed 10、20、27 days after using PTZ.Results: There was significant difference in behavior between group P and TP after 4 weeks.Expression of GFAP increased in group P and TP(P0.05);and continuously increased along with time.and the expression in P group is more higher than in group TP at different time-points(P0.05).Conclusion: The increasing of Expression of GFAP in hippocampus indicates that reactive gliosis occurs and neural plasticity maybe an important mechanism of epileptogenesis in the process of PTZ kindling rats.Expression of GFAP is inhibited markedly by TPM,which indicates that TPM can influence expression of GFAP and may influence nerve plasticity.
