Abstract
Protoplast culture and plant regeneration are the basic techniques for genetic engineering of plants.Protoplast sources,enzyme isolation conditions,medium compositions and culture methods etc.have obvious effects on the protoplast culture of plants.The factors affecting plant regeneration from protoplasts of tobacco were described comprehensively here.And the potential application of protoplast culture technique in plant breeding was also discussed.
