Abstract
Objective To investigate the induction of apoptosis and generation of reactive oxygen species(ROS) of human liver cell L-02 in vitro by triptolide.Methods L-02 cells were cultured in vitro and the cytotoxic effects were detected by MTT.ROS was determined by FCM with the fluorescent probes H2DCFDA.The activity of SOD,LDH,and MDA concentration was detected.Results Triptolide markedly promoted ROS accumulation,and it accumulated to th maximun value when cultured for 12 h(P0.01).Triptolide significantly increased MDA concentration,LDH release and decreased SOD activity.Conclusion Triptolide induces the apoptosis of L-02 cells which is associated with the induction of ROS generation.
