Soil-supporting Rooting Culture and Lump-transplanting of Cotoneaster hjelmqvistii Test-tube Plantlets

Abstract

[Objective] The purpose was to improve the rooting and transplanting techniques of Cotoneaster hjelmqvistii test-tube plantlets.[Method] With clay loam as medium supporter,the liquid MS medium without macro-element and with 0.75 mg/L IBA and 15 g/L sucrose was selected for rooting culture of stem segments of C.hjelmqvistii test-tube plantlets.With agar-supporting culture as CK,the rooting conditions of C.hjelmqvistii test-tube plantlets cultured by different methods and their lump-transplanting effects were studied.[Result] The rooting rate,root length,root number and leaf number of C.hjelmqvistii test-tube plantlets in treatment of soil-supporting culture(being 95.0%,54.3 mm,2.8 and 7.0 resp)were significantly higher than those of agar-supporting culture.The C.hjelmqvistii test-tube plantlets in treatment of soil-supporting culture and agar-supporting culture all had root hair.When the rooted C.hjelmqvistii test-tube plantlets in treatment of soil-supporting culture were transferred with lump into nutrient body after open-bottle acclimation for 21 d,their survival rate could reach 96.7% under the condition without spray and film covering and with low relative air humidity of 50%.[Conclusion] With clay loam as medium supporter,the rooting and lump-transplanting effects of C.hjelmqvistii test-tube plantlets could be enhanced significantly.
