Determination of the content of protocatechuic acid in rhizome of Alpinia Officinarum by HPLC

Abstract

Objective To develop a HPLC method for the determination of protocatechuic acid in the herbal of Alpinia officinarum Hance from different places.Methods A Diamonsil C18 column(200 mm×4.6 mm,5 μm)was used with a mobile phase consisting of methanol-0.05%(w)phosphoric acid(V∶V=12∶88)at a flow rate of 1.0 mL·min-1.The detection wavelength was set at 258 nm,and the column temperature was 35 ℃.Results The liner range for protocatechuic acid was 0.902-14.432 mg·L-1.The average recovery for protocatechuic acid was 99.2%(RSD=1.8%,n=9).Conclusions The method can be used to control the quality of Alpinia officinarum Hance,which provides the advantages of quickness,simplicity and repeatability.
