Abstract

Objective:To establish a method for the simultaneous determination of myristic acid,palmitic acid,stearic acid,oleic acid and linoleic acid in wood frogs.Methods:The sample was determined with PEG-20M capillary column(30 m×0.25 mm,0.25 μm) by GC.The detector was FID;the initial teperature was 200 ℃ matained 2 min,the program was 200 ℃ programmed to 230 ℃ at 4 ℃·min-1 for 8 min;the injector temperature was 260 ℃;the detector temperature was 280 ℃;the flow rate was 0.3 mL·min-1;split injection was conducted with split radio of 20∶1.Results:The linear response ranges of myristic acid,palmitic acid,stearic acid,oleic acid and linoleic acid were 0.015-0.152 mg·mL-1(r=0.9994),0.106-1.056 mg·mL-1(r=0.9993),0.020-0.203 mg·mL-1(r=0.9991),0.133-1.332 mg·mL-1(r=0.9995) and 0.055-0.545 mg·mL-1(r=0.9992),respectively(n=6);the mean recoveries(n=9) of myristic acid,palmitic acid,stearic acid,oleic acid and linoleic acid were 97.4%,97.6%,97.9%,97.4% and 96.0%,respectively.Conclusion:The method is simple and accurate for the determination of myristic acid,palmitic acid,stearic acid,oleic acid and linoleic acid in spawn of wood frogs.
